Expression Library and Protein Pool

The P. furiosus in vitro expression library was the same as reported in [4]. In summary, P. furiosus genome DNA was digested into 7kb on average and integrated into pRSET A vector and transformed into E.coli strain DH 5$\alpha$ . The P. furiosus proteins were collected from cell culture centrifugation product after sonication and heat treatment to remove E.coli endogenous proteins. This library contains 700 protein pool, with each pool covers the expression of about 7kb P. furiosus genomic DNA, which means about 7 proteins in each lysate.
