It has been known that many RNA binding protein bind
different RNA oligo structure such as double stranded RNA, single stranded RNA,
loop and bulge. Based on this, we designed the MPOR27 for high efficiently
minding RNA-binding proteins. In this work, we systematically screened about 700
pool mixtures for novel RNA-binding proteins using a multi-structure oligo RNA
(MPOR-27). It is named multi-structure because it consists of 4
different secondary structure : ss (single stranded), ds (double stranded),
bulge and a loop. For any RNA-binding protein, even if it can only recognize one
of the four structures, it shall be able to bind MPOR-27 steadily. With 4
different structures, which cover the current known protein-binding domain
structures, MPOR-27 shall be able to find all the RNA-binding proteins in any
protein pool under optimum binding conditions.
